Jerry,
I don't have info on public transport, but that is a great trip you are planning. I've been up there three times to all those places (don't forget hell fire pass and the natural hot spring baths on the way to three pagoda pass) in my own vehicle and thoroughly enjoyed the trip each time.

My brain is like an Internet browser; 12 tabs are open and 5 of them are not responding, there's a GIF playing in an endless loop,... and where is that annoying music coming from?

If you go slightly south of the city centre - walking distance - on the river and just up from the small bar area - there are a number of decent guesthouses that are in the 400-500 THB range.
I stayed at one once (sorry can't remember the name) and it was fine. Fan room, clean and western bathroom. Friendly Thai hosts and good food.
I'm sure that anywhere you stay will be able to organise tours for you or alternatively you might find slightly cheaper deals if you talk to operators direct.
Public transport? I think the train takes some time. I've got no idea about buses.

Hi Jerrie,
Kanchanaburi is my favourite place for a bit of R&R, but I don't think that public transport (i.e. buses & trains) go directly there. If you get the bus, you'll have to get off it somewhere just past Big C in Petchaburi, and then catch another bus to Kanchanaburi. This is info. from a student, so it's rather sketchy and possibly unreliable...
The alternative is to get a train to Nakhon Pathom - then catch another one to Kanchanaburi. It could be a long journey if the timing isn't right, and if you're limited on time you'd be wasting hours. Maybe a taxi is your best bet?
When you get there I'd highly recommend visiting one of the War Cemeteries and the JEATH museum - you can do both of these in a long-tailed boat (which is fun!). Lunch in the restaurant on the river just by the bridge is also good value for money, and you get a fantastic view of the bridge.
Enjoy your trip.
